I Am An Employer - Question Regarding Unemployment Insurance?

I am not a new business, but now have employees. Ive hired a local payroll company, and I am all set with disability insurance, and workmens comp insurance. I now have a question as Ive just found out I need to file form NYS100 for unemployment insurance. Is this something else Im going to have to pay now or is it just a form you file and then have to pay if you fire or lay off an employee? I only have 5 employees and all are part time. Can someone please explain how this works I am so confused and I thought I had everything set and now I realize I need this too.
Thanks in advance for any advice you can give. Im in NY if that matters.

Unemployment is something you'll have to pay into while your employees are working.
